Fixation d''un anticorps anti-lysozyme dans des cellules de mollusques bivalves ArchiMer
Auffret, Mathieu; Diouris, M; Herry, A.
A polyclonal rabbit anti-human lysozyme (Dako) was applied on histological sections from bivalve mollusc and revealed with a mouse anti-rabbit IgG fluorescein conjugated antibody to localise cellular lysozyme in tissues where its activity had previously detected by enzymatic techniques. Labelling occurred in the cytoplasm of epithelial cells covering the body. This is in agreement with the intracellular distribution of this enzyme. In the Lucinid Lucinella divaricata gills, the cytoplasm of bacteriocytes was also labelled. This suggests that antibacterial activity occurred in these cells. Control sections indicate that fluorescence was a consequence of true primary antibody fixation. However, its specificity should be verified by electrophoretical...

A STUDY OF ASSOCIATION BETWEEN ELISA RESPONSE TO INFECTIOUS DISEASE AGENTS AND CALVING INTERVAL IN CATTLE IN THE TROPICS OF MÉXICO Ciência Rural
Barajas-Rojas,José Alfonso; Riemann,Hans; Franti,Charles.
Forty eight cows of Holstein-Zebu crosses with at least one completed pregnancy were tested for antibodies to 20 disease agents. The testing was performed within one month (average two weeks) after calving when the IgG level constantly reaches its lowest value during the cows reproductive cycle. The results indicate that increasing levels of IgG against Leptospira interrogans serovar hardjo and Mycobacterium paratuberculosis may be associated with an increased calving interval. Increase in calving intervals could not be explained by age of cows or their genotypic characteristics.

Baraldi,Thaís Gasparini; Almeida,Henrique Meiroz de Souza; Morais,Amanda Bonalume Cordeiro de; Storino,Gabriel Yuri; Montassier,Hélio José; Motta,Rodrigo Garcia; Ribeiro,Márcio Garcia; Oliveira,Luís Guilherme de.
ABSTRACT: Family Tayassuidae in the suborder Suina include two species of peccaries in Brazil: the white-lipped peccary (Tayassu pecari) and the collared peccary (Pecari tajacu). These animals share common pathogens with domestic swine (Sus scrofa); however, their role as potential carrier remains unclear. This study focused on detecting the prevalence of influenza A antibodies in Tayassu pecari and Pecari tajacu from commercial rearing farms from two states in Brazil. A set of 50 blood samples from Pecari tajacu and 55 from Tayassu pecari were analyzed using a commercial indirect ELISA in order to investigate anti influenza A antibodies. Pecari tajacu samples presented 22% (11/50) of seropositivity for the virus. Serological surveillance is an important...

Comparison of dot-ELISA and standard ELISA for detection of Neisseria meningitidis outer membrane complex-specific antibodies BJID
Belo,Elza FT; Farhat,Calil K; Gaspari,Elizabeth N De.
Dot-ELISA using the outer membrane complex antigens of Neisseria meningitidis as a target was standardized for rapid detection of meningococcal-specific antibodies in human serum. We investigated the level of meningococcal-specific IgG, IgA, and IgM in serum using dot-ELISA with outer membrane antigens prepared from Neisseria meningitidis serotype B:4.19:P1.15,3,7,9 (a strain isolated from a Brazilian epidemic). The dot-ELISA is based on the same principles as the standard ELISA and is useful for detection of anti-N. meningitidis B antibodies in serum of patients with meningococcal infections. For the assay, outer membrane complexes (OMCs) were absorbed by nitrocellulose membrane and blocked with a 5% skim milk solution. Serum samples were drawn upon...

TOXIC SHOCK SYNDROME J. Venom. Anim. Toxins
BERGDOLL,M. S..
Toxic shock syndrome is a staphylococcal disease caused by toxins produced by the staphylococci, toxic shock syndrome toxin-1 and enterotoxin B. The disease results from staphylococci growing in the vagina with the use of tampons during menstruation, primarily in young women. However, any staphylococcal infection can result in toxic shock syndrome if the staphylococci produce the appropriate toxins and the individual has no antibodies to the toxins. The symptoms can be quite severe, with high fever, low blood pressure, diffuse macular erythroderma, orthostatic dizziness, vomiting and or diarrhea at the onset, severe myalgia, peeling of the skin of the palms of the hands and the soles of the feet after seven to ten days, and death in some cases. The disease...

Production of specific polyclonal antibodies anti-human factor IX BABT
Chaves,Daniel Gonçalves; Rodrigues,Cibele Velloso; Ferreira,Wanderley Almeida; Siqueira,Pollyana Fantini Miranda; Duarte,Clara Guerra; Carneiro-Proietti,Anna Bárbara de Freitas; Santoro,Marcelo Matos.
In this work, polyclonal antibodies anti-human Factor IX were produced in New Zealand rabbits by immunization with commercial pure human FIX (hFIX) (Octanyne®, Octapharma, USA). The serum containing immunoglobulins anti-hFIX was useful to detect hFIX antigen in human plasma fractions submitted to anionic exchange chromatographic process and with a large yield. Immunoassays (ELISA) using bovine serum albumin, trypsin and peptides generated by cleavage assays with trypsin as digestion enzyme was performed and revealed adequate specificity of the polyclonal antibodies produced.

Sera from chronic chagasic patients depress cardiac electrogenesis and conduction BJMBR
Costa,P.C.S.; Fortes,F.S.A.; Machado,A.B.; Almeida,N.A.C.; Olivares,E.L.; Cabral,P.R.; Pedrosa,R.C.; Goldenberg,R.C.; Campos-de-Carvalho,A.C.; Masuda,M.O..
We report results obtained with sera from 58 chronic chagasic patients that were evaluated for effects on heart rate and atrioventricular (AV) conduction in isolated rabbit hearts and screened for the presence of muscarinic and beta-adrenergic activity. We show that sera from 26 patients decreased heart rate, while 10 increased it and 22 had no effect. Additionally, sera from 20 of the 58 patients blocked AV conduction. Muscarinic activation seems to be involved in both effects, but is not the only mechanism, since atropine did not antagonize the decrease in heart rate in 23% of sera or AV block in 40%. Sera from patients with complex arrhythmias were significantly more effective in depressing both heart rate and AV conduction. Sera that induce increases...

Antibodies against pseudorabies virus in feral swine in southeast Brazil Arq. Bras. Med. Vet. Zootec.
Cunha,E.M.S.; Nassar,A.F.C.; Lara,M.C.C.S.H.; Bersano,J.G.; Villalobos,E.M.C.; Oliveira,J.C.F..
Serum samples collected from 358 wild boars (Sus scrofa) in breeding farms in São Paulo, southeast Brazil, from 1998 to 2001, were tested for antibodies against pseudorabies virus (PRV) by means of serum neutralization (SN) and enzyme-linked immunobsorbent assay (ELISA). Seropositive animals were detected in three of seven herds analyzed. Overall seroprevalence as assessed by SN was 30.7%, ranging from 25.2% to 100% for the herds that presented seropositive animals. Indirect ELISA detected lower seroprevalence (19.3%). Sensitivity and specificity of ELISA were equal to 57.3% and 97.6%, respectively. Agreement was equal to 85.2% (P<0.0001). These results showed that PRV infections occurred in farmed feral swine in southeast Brazil, and affect...

DNA vaccines for viral diseases BJMBR
Donnelly,J.J.; Ulmer,J.B..
DNA plasmids encoding foreign proteins may be used as immunogens by direct intramuscular injection alone, or with various adjuvants and excipients, or by delivery of DNA-coated gold particles to the epidermis through biolistic immunization. Antibody, helper T lymphocyte, and cytotoxic T lymphocyte (CTL) responses have been induced in laboratory and domesticated animals by these methods. In a number of animal models, immune responses induced by DNA vaccination have been shown to be protective against challenge with various infectious agents. Immunization by injection of plasmids encoding foreign proteins has been used successfully as a research tool. This review summarizes the types of DNA vaccine vectors in common use, the immune responses and protective...

Bacterial infections occur worldwide and are a major public health problem. Among pathogens, Staphylococcus aureus is the main causative agent of bacterial diseases in the world. This study aimed to evaluate which components of the immune system could act protectively against a S. aureus infection in intradermally immunized mice. C57BL/6 and A/j mice were immunized intradermally with S. aureus inactivated by heat and then challenged with viable strains in an air pouch model. At 6, 12, and 24 h after the challenge, euthanasia was performed, and the cellular profile of the inflammatory infiltrate, cytokines, and the bacterial load were evaluated in the air pouch lavages. Immunized mice demonstrated that the intradermal immunization with S. aureus promoted...

Expression of Grapevine leafroll-associated virus 3 coat protein gene in Escherichia coli and production of polyclonal antibodies Trop. Plant Pathol.
Fajardo,Thor V.M.; Barros,Danielle R.; Nickel,Osmar; Kuhn,Gilmar B.; Zerbini,F. Murilo.
Grapevine leafroll-associated virus 3 (GLRaV-3), the main viral species of the grapevine leafroll complex, causes yield and quality reduction in grapes (Vitis spp.). The coat protein gene was RT-PCR-amplified from total RNA extracted from infected grapevine leaves and the amplified fragment was cloned and completely sequenced. The fragment was subsequently subcloned into the pRSET-C expression vector. The recombinant plasmid was used to transform Escherichia coli BL21:DE3 and express the capsid protein. The coat protein, fused to a 6 His-tag, was purified by affinity chromatography using an Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE and Western blot. The in vitro-expressed protein was quantified and used for rabbit...

Evaluation of antibodies production against Borrelia burgdorferi in cattle submitted to rBm86 protein Boophilus microplus tick immunization and associated challenges influence BJM
Ishikawa,Márcia Mayumi; Fonseca,Adivaldo Henrique; Yoshinari,Natalino Hajime; Osório,Ana Luiza Alves Rosa.
IgG antibodies production against Borrelia burgdorferi in immunized cattle with rBm86 protein from Boophilus microplus was evaluated as well as the influence of the association between immunizations and stress through indirect ELISA test during one year. In the present study there was no influence of the isolated challenged used on the production of IgG antibodies against B. burgdorferi. The rBm86 immunogen did not cause significant oscillation in the production of IgG antibodies against B. burgdorferi capable to interfere in the serological results for Lyme Borreliosis in cattle. This study demonstrated the possibility of transitory changes in the production of antibodies after the association of vaccine stimuli and stress, emphasizing the necessity of...
